About TB-500
TB-500 is a synthetic peptide that reproduces the active region of thymosin beta-4 (Tβ4), a naturally occurring 43-amino-acid protein present in virtually all human cells. Commercial TB-500 is usually a short fragment (an N-acetylated region carrying the active LKKTETQ motif) with a molecular mass of roughly 1.9 kDa. It is supplied as a lyophilized powder and is intended for laboratory research use only.
How it works
TB-500's key mechanism relates to actin regulation: the active LKKTETQ sequence binds G-actin monomers, influencing actin cytoskeleton polymerization and cell motility, which in research is associated with the migration of reparative cells toward the site of injury. In parallel, preclinical models describe stimulation of angiogenesis through increased VEGF expression and integrin signaling — that is, the formation of new blood vessels. Modulation of inflammation (the NF-κB pathway) and reduced fibrosis via effects on TGF-β-dependent collagen deposition have additionally been reported.
What's studied / benefits
- Accelerated cell migration to the injury site and regulation of the actin cytoskeleton in healing models
- Stimulation of angiogenesis (formation of new blood vessels) through increased VEGF expression
- Reduced fibrosis and scarring through effects on TGF-β-dependent collagen deposition
- Modulation of inflammatory processes via the NF-κB pathway in preclinical studies
- The strongest preclinical signals are in the context of tendon, ligament, muscle and cardiac tissue recovery
Research applications
- Research into wound healing and soft tissue recovery
- Study of tendon and ligament repair (tendinopathy models)
- Research into cardiac tissue regeneration and fibrosis reduction
- Study of angiogenesis, neuroprotection and other regenerative processes
Of interest to: TB-500 typically appeals to researchers in regenerative biology and tissue repair, as well as biohackers and the athletic audience focused on post-exercise recovery topics (important: in professional sport the substance is banned by WADA).
