About GLP-3 / RT

RT (code name LY3437943) is a synthetic 39-amino-acid peptide developed by Eli Lilly. It is informally referred to as "GLP-3" because it is a triple agonist of three receptors at once — GLP-1, GIP and glucagon — which sets it apart from single (GLP-1) and dual (GLP-1/GIP) molecules. It is supplied strictly for laboratory research and is not an approved medicinal product.

How it works

The molecule simultaneously activates three receptors: GLP-1 (signals satiety to the hypothalamus and slows gastric emptying), GIP (enhances glucose-dependent insulin secretion and affects fat metabolism) and glucagon — and it is this last component that is the key distinction. Glucagon receptor agonism increases energy expenditure through thermogenesis and stimulates lipolysis and hepatic fat oxidation. As a result, studies observe a two-way effect on adipose tissue: reduced calorie intake together with increased energy expenditure.

What's studied / benefits

  • In a Phase 2 clinical trial (NEJM, 2023), the average reduction in body weight reached 24.2% at week 48 at a dose of 12 mg/week, with a plateau not yet reached
  • In Phase 3 (Eli Lilly report, 2025), an average body weight reduction of up to ~28.7% over 68 weeks was reported
  • In a Phase 2a study in MASLD (metabolic dysfunction-associated steatotic liver disease, Nature Medicine, 2024), a pronounced reduction in liver fat content — of more than 80% — was recorded
  • In people with type 2 diabetes (Lancet, 2023), a clinically meaningful improvement in glycemic control was observed along with weight loss
  • Studies noted a concomitant improvement in metabolic markers — triglycerides and blood pressure

Research applications

  • Research into obesity and excess body weight as the molecule with the highest weight-loss potential in its class
  • Study of metabolic dysfunction-associated steatotic liver disease (MASLD/NAFLD) and reduction of liver fat
  • Research into glycemic control in type 2 diabetes
  • Comparative study of triple agonism (GLP-1/GIP/glucagon) versus single and dual agonists

Of interest to: Of greatest interest to metabolism and obesity researchers, as well as to biohackers and the sports community who follow cutting-edge compounds in this class — given its record-setting research potential to affect body weight and fat metabolism.
